Introduction to Masterclass

scRNA and CRISPR technology are transforming the landscape of genomic research. By combining scRNA & CRISPR, scientists can achieve unprecedented precision in studying gene function at the single-cell level, unveiling new insights into cellular diversity and disease mechanisms.

scRNA sequencing allows researchers to analyze gene expression in individual cells, providing a detailed snapshot of cellular states and transitions. This high-resolution approach is essential for understanding the complexity of tissues and identifying rare cell populations that might be overlooked in bulk RNA sequencing. Meanwhile, CRISPR technology offers a powerful method for precise genome editing, enabling targeted modifications to DNA sequences. When integrated with scRNA and CRISPR can be used to systematically perturb genes and observe the resulting changes in single-cell gene expression.

The synergy between scRNA & CRISPR has led to the development of innovative techniques such as CRISPR-based lineage tracing and perturb-seq. These methods enable the mapping of cellular lineages and the functional interrogation of genes in a high-throughput manner. This approach facilitates the identification of gene regulatory networks and the discovery of novel gene functions.

Applications of ScRNA & CRISPR

In this section we are discussing some of the potential areas of application of ScRNA and CRISPR & how it can be helpful in these areas to carryout analysis.

scRNA and CRISPR are used together to systematically perturb genes and assess their functions at the single-cell level.

In oncology, scRNA & CRISPR are employed to study tumor heterogeneity, identify cancer driver genes, and understand resistance mechanisms to therapies.

Researchers use scRNA & CRISPR to investigate the gene regulatory mechanisms underlying cell differentiation and tissue development.

scRNA sequencing combined with CRISPR technology is utilized to dissect the complexity of the immune system, particularly in identifying gene functions in immune cell subsets.

In neurobiology, scRNA & CRISPR are used to study gene functions in neural cells and understand the molecular basis of neurological disorders.

Different Types of Analysis

Majorly, there are 3 different types of analysis using ScRNA and CRISPR so below we have tried to list them down to help you take better decision if this masterclass will be relevant for your learning and be helpful in your intended area of research.

Perturb-Seq Analysis

In Perturb-Seq, CRISPR is used to introduce targeted gene disruptions across a population of cells. scRNA sequencing is then performed to measure the transcriptional changes resulting from these perturbations.

Lineage Tracing & Fate Mapping

CRISPR is utilized to introduce unique genetic barcodes or edit specific lineage markers in single cells. scRNA sequencing is then used to capture the transcriptomes of these cells over time to trace cell lineage & map cell fate decision.

CRISPRa-Seq

CRISPR activation (CRISPRa) techniques, which use dead Cas9 (dCas9) fused to transcriptional activators, are employed to upregulate target genes in individual cells. scRNA sequencing is then performed to assess the resulting changes
